Anti-inflammatory effect of Terrein in periodontal Ligament cells

Abstract

|
Objective: Terrein was isolated from a microorganism Penicillium species 20135, and Terrein has a anti-oxidant activity and anti-inflammatory activity in human dental pulp cells. However its anti-inflammatory effects are unknown in other types of oral cells. The purpose of this study is to determine the anti-inflammatory effects of Terrein in periodontal ligament cells.
Material and Methods: Human periodontal ligament cells(PDL) obtained from permanent and noncarious teeth were cultured and only third to eighth passages were used. PDL cells were exposed to various concentration of LPS and Terrein. Anti-inflammatory potential of Terrein was checked with expression of MMPs, intercellular adhesion molecule-1(ICAM-1), and vascular cell adhesion molecule-1(VCAM-1).
Results: PDL cells exposed to LPS(1 - 10§¶/§¢) revealed elevated levels of MMP-2, MMP-9, ICAM-1 and VCAM-1. LPS-induced expression of MMP-2, MMP-9, ICAM-1 and VCAM-1 were significantly inhibited by Terrein in a dose and time dependent manner.
Conclusion: These result offers us the possible use of Terrein as an anti-inflammatory agent in periodontitis.
